چکیده

Circadian variations are observed in many physiological processes, and alterations in these oscillations are closely linked to mood disorders, metabolic abnormalities and cardiovascular complications. The nuclear receptor Rev-erba is a crucial component of the clock mechanism. It is an important metabolic regulator and a target for lithium, a drug used in the treatment of bipolar disorders. The recent identification of natural (heme) and synthetic ligands for Rev-erba suggests that pharmacological modulation of Rev-erba activity may be a potential therapeutic option in the treatment of circadian disorders including circadian-related metabolic and cardiovascular diseases. Integrated Systems and Technologies: Mathematical Oncology A Circadian Clock Transcription Model for the Personalization of Cancer Chronotherapy

Circadian timing of anticancer medications has improved treatment tolerability and efficacy several fold, yet with intersubject variability. Using three C57BL/6-based mouse strains of both sexes, we identified three chronotoxicity classes with distinct circadian toxicity patterns of irinotecan, a topoisomerase I inhibitor active against colorectal cancer. Liver and colon circadian 24-hour expre...

Molecular Mechanism Regulating 24-Hour Rhythm of Dopamine D3 Receptor Expression in Mouse Ventral Striatum s

The dopamine D3 receptor (DRD3) in the ventral striatum is thought to influence motivation and motor functions. Although the expression of DRD3 in the ventral striatum has been shown to exhibit 24-hour variations, the mechanisms underlying the variation remain obscure. Here, we demonstrated that molecular components of the circadian clock act as regulators that control the 24-hour variation in ...

The Nuclear Receptor REV-ERBa Regulates Fabp7 and Modulates Adult Hippocampal Neurogenesis

The function of the nuclear receptor Rev-erba (Nr1d1) in the brain is, apart from its role in the circadian clock mechanism, unknown. Therefore, we compared gene expression profiles in the brain between wild-type and Rev-erba knock-out (KO) animals. We identified fatty acid binding protein 7 (Fabp7, Blbp) as a direct target of repression by REV-ERBa. Loss of Reverba manifested in memory and moo...

Impact of Circadian Nuclear Receptor REV-ERBα on Midbrain Dopamine Production and Mood Regulation

The circadian nature of mood and its dysfunction in affective disorders is well recognized, but the underlying molecular mechanisms are still unclear. Here, we show that the circadian nuclear receptor REV-ERBα, which is associated with bipolar disorder, impacts midbrain dopamine production and mood-related behavior in mice. Genetic deletion of the Rev-erbα gene or pharmacological inhibition of ...
